Thrombosis is associated with the coagulation cascade in blood triggered by sudden plaque disruption in heart diseases such as acute myocardial infarction and ischemic stroke. GPIIIb/IIa has been demonstrated to play an important role in mediating platelet activation and clot retraction. Within GPIIIb/IIa signaling, I particularly interest in the correlation between Src/Syk tyrosine kinases and cysteine protease calpains in regulating the platelet signaling cascade. By using a tyrosine kinase inhibitor, calpains inhibitor alone and in combination, I am studying how platelets function in adhesion, spreading and aggregation. The results will help to understand how these two classes of enzymes interact in platelet signaling, and the additive effect of the inhibitor combination may suggest a potential modality for preventing platelet-induced thrombosis.
