My research project is investigating the mechanisms of G protein signaling pathways in platelets and other mammalian cells. Briefly, the switch regions of G protein Gα subunits activate downstream cell signaling events. Gα13 Switch Region 2 forms a calcium dependent bi-molecular complex with the head domain of talin. Binding of the Gα13 to the talin head domain promotes αIIbß3 integrin activation. These studies provide a new paradigm for inside-out signaling and αIIbß3 integrin activation in platelets, as well as the mechanism of cell migration and adhesion abnormalities in cancer cells.
